I think you will have to arrange it all in advance. While streets are wheelchair accessible, many of the shops and museums are not.

If you like history then you need at least a couple of days in Williamsburg. You can do Jamestown and Yorktown in one day each.

Jamestown - yes. Yorktown - bus plus taxi. Bus leaves from and returns to the Williamsburg Transportation Center. No actual experience traveling to either destination by bus.
